Determination of Olprinone and Related Materials by RP-LC

Abstract
Olprinone hydrochloride is an inhibitor of cyclic nucleotide phosphodiesterase III. A reverse phase liquid chromatography method was established for the determination of olprinone and related materials (4 synthetic intermediates). An Agela Venusil ASB C18 reversed phase column (4.6 mm ID × 25 cm, 5 μm particle size) with a flow rate of 1.0 mL/min at 215 nm was used. The mobile phase consisting of 0.01 mol/L dipotassium hydrogen phosphate and methanol in the ratio of 92:8 (pH = 2.5). The calibration curve for olprinone was linear in the concentration range of 40-200 μg/mL (r = 0.9993, n = 6). The average recovery was 99.56 % with RSD of 0.83 %. By using proposed HPLC method, olprinone and impurities were well-separated. This method is rapid, sensitive and practical.
